Overview

[3H]-cholesterol bound to albumin is a radiolabeled diagnostic tracer developed by the University of Pennsylvania to evaluate reverse cholesterol transport (RCT) in vivo. The tracer consists of tritiated cholesterol associated with human serum albumin, administered intravenously as a bolus. It is used in clinical validation studies to assess the rate and efficiency of cholesterol movement from the plasma into lipoprotein fractions and eventually into feces. This method is specifically designed to study patients with monogenic disorders affecting high-density lipoprotein (HDL) metabolism, including Tangier disease (caused by ABCA1 mutations), lecithin-cholesterol acyltransferase (LCAT) deficiency, and cholesteryl ester transfer protein (CETP) deficiency. By tracking the radioactivity of the 3H-tracer, researchers can quantify the RCT pathway, providing insights into the pathophysiology of these rare genetic conditions and the potential impact of therapeutic interventions on cholesterol clearance.
